Selectivity of cations and nonelectrolytes for acetylcholine-activated channels in cultured muscle cells
The selectivity of acetylcholine (A-Ch)-activated channels for alkali cations, organic cations, and nonelectrolytes in cultured muscle cells has been studied. To test the effect of size, charge, and hydrogen-binding capacity of permeant molecules on their permeability, we have obtained the selectivity sequences of alkali cations, compared the permeability of pairs of permeant molecules with sim...
متن کاملIon conductance and selectivity of single calcium-activated potassium channels in cultured rat muscle
The conductance and selectivity of the Ca-activated K channel in cultured rat muscle was studied. Shifts in the reversal potential of single channel currents when various cations were substituted for Ki+ were used with the Goldman-Hodgkin-Katz equation to calculate relative permeabilities. The selectivity was Tl+ greater than K+ greater than Rb+ greater than NH4+, with permeability ratios of 1....
متن کاملTranscripts for the acetylcholine receptor and acetylcholine esterase show distribution differences in cultured chick muscle cells
In situ hybridization of chick cultured muscle cells using exonic DNA probes for both AChR alpha-sub-unit and the catalytic subunit of AChE, revealed major differences in the distribution of label both over nuclei and in their surrounding cytoplasm, although some overlap in these distributions exists. For the AChR alpha-subunit there is a highly skewed distribution of labeled nuclei, with 35% o...
متن کاملDevelopment of acetylcholine receptor clusters on cultured muscle cells.
Clusters of acetylcholine receptors were formed in the absence of neurons during the development of cultured chick-embryo skeletal muscle cells. The average concentration of receptors in clusters was estimated to be 9000 per mum(2). At other regions of the cell, receptor concentration was 900 per mum(2). The possibility that receptor clusters may participate in synapse formation is suggested.
متن کاملSlow permeation of organic cations in acetylcholine receptor channels
Block, permeation, and agonist action of small organic amine compounds were studied in acetylcholine receptor (AChR) channels. Single channel conductances were calculated from fluctuation analysis at the frog neuromuscular junction and measured by patch clamp of cultured rat myotubes. The conductance was depressed by a few millimolar external dimethylammonium, arginine, dimethyldiethanolammoniu...
